ZL-6201 (LRRC15 ADC)

Overview

ZL-6201 is an internally developed, potential first- and best-in-class investigational antibody-drug conjugate (ADC) that demonstrates high affinity and specificity for LRRC15.  This target is heavily upregulated in the sarcomas and cancer-associated fibroblasts (CAFs) of multiple solid tumors. By leveraging the next-generation TMALIN® ADC technology platform, ZL-6201 is specifically engineered to overcome the limitations of first-generation ADCs, aiming to maximize targeted efficacy while mitigating systemic toxicities. 

ZL-6201 efficiently targets LRRC15 expressed on both tumor cells and CAFs. Upon internalization and payload release, the ADC drives cell death in both directly targeted cells and neighboring tumor cells within the tumor microenvironment through a potent bystander effect. Robust in vitro and in vivo preclinical studies have demonstrated broad efficacy in models where LRRC15 is present either directly on the cancer cells or within the stroma from CAFs. Zai Lab has initiated a global Phase 1 clinical trial evaluating ZL-6201 in patients with sarcoma and other epithelial tumors.
